Studies on Tissue Culture of Rickettsia

Abstract
|

|
Zinsser´s agar slant tissue culture method has made Rickettsial growth successful to get much yield and produce vaccine, however it was not easy to reach constant results f or the followers as it was not mentioned in technical details. Author have tried to get through governing technical points on rickettsial culture in order to prevent fails and could clear several hard points out as ´follows;
1. The size of minced tissue, as well as mincing skill were the key factors for the rickettsial gro wth and when they were adequate, PH of media didn´t -move quickly and self softening process´ of planted tissue pieces up to translucent mucoid droplets appeared regular
2. Interval of transfer period was better choose 6 days than 8-9 days when the rickettsial yield´ reach to its maximum.
3. Possible chances of bacterial contamination were the most exclusive facors for rickettsial tissue culture and simplified manipulation must be sought and planed through out the culture process of rickettsia.
4. Ox serum Was substituted for horse serum´for the-sake of accesibility.
